Trisomy 21-induced metabolite alterations drive cellular injury in Down syndrome: mechanistic insights and therapeutic potential

This study used isogenic induced pluripotent stem cells from Down syndrome patients to investigate how the extra chromosome 21 disrupts cellular metabolism and causes DNA damage, identifying two key metabolites (5-oxo-ETE and Calcitriol) with opposite effects on cell health. Notably, Calcitriol supplementation showed protective effects against DNA damage and improved cardiogenesis, suggesting a potential therapeutic avenue for addressing Down syndrome complications.
